Tubulin Tracker Deep Red provides deep-red/far-red fluorescence when bound to polymerized tubulin in live cells. Tubulin Tracker Deep Red is based on Docetaxel conjugated with a bright, photostable deep-red fluorophore. Docetaxel belongs to the family of cytoskeletal drugs that target tubulin. Tubulin Tracker Deep Red absorbs and emits optimally at 652 nm and 669 nm, respectively and can be visualized with standard Cy5 filter settings using almost any fluorescent imaging instrument. It can be multiplexed with blue, green, orange, red, and near-IR fluorophores.

•1 vial Tubulin Tracker Deep Red (Add 60 μL/vial of anhydrous DMSO to make a 1000X (1mM) stock solution.  Dissolve stock solution 1:1000 in cell imaging solution to make 1X (1 μM) staining solution.)
•1 x 77 mg Probenecid (Dissolve vial content in 1 mL of live cell compatible buffer to make a 100X stock solution.)

Store at ≤–20°C after recieving.
Desiccate and protect from light.
When stored as directed, product is stable for at least 6 months.

Frequently asked questions (FAQs)

With Tubulin Tracker Deep Red and Tubulin Tracker Green, can I fix samples after labeling?

No. The labeling is not retained upon fixation. Imaging of the samples should be done only on live cells. To limit photobleaching while viewing live cells, use ProLong Live Antifade Reagent, for live cell imaging (Cat No. P36974 and P36975).

How is Docetaxel in Tubulin Tracker Deep Red different from Taxol in Tubulin Tracker Green? Are there any differences in the binding or cytotoxicity between the two reagents?

Docetaxel has a slightly higher binding affinity than Taxol and exhibits longer retention time due to better uptake and slower efflux, but for use in labeling cultured cells, these differences should be minimal. Is Tubulin Tracker Deep Red acetylated? Does it require intracellular esterase activity to make the reagent active?

Tubulin Tracker Deep Red is not acetylated. It is inherently cell permeable and is not modified further by any cellular enzymatic activity to fluoresce or bind to actin.

Do I need to use Probenecid when using Tubulin Tracker Deep Red?

The use of Probenecid is optional. Its requirement is dependent upon the cell type. Some cell types actively efflux the reagent out of the cells within a short time, limiting detection, other cell types do not exhibit this activity or exhibit very limited activity.
