ReadyProbes 2.5% Normal Horse Serum is a ready-to-use non-specific blocker for use in immunohistochemistry (IHC), immunocytochemistry (ICC), and immunofluorescence (IF) experiments, usually when a horse or donkey secondary antibody is used. Incubating cells/tissue with diluted serum from the same or similar species as the host of the secondary antibody typically reduces non-specific binding. Simply cover the tissue section or cells with a few drops of ReadyProbes 2.5% Normal Horse Serum and incubate for 30–60 minutes at room temperature. Ready Probes 2.5% Normal Horse Serum can also be used to dilute primary and secondary antibodies.
Ready Probes 2.5% Normal Horse Serum is provided as a ready-to-use reagent with no further dilution or preparation required. The serum is collected from healthy adult animals and the heat-treated, centrifuged, and filtered. It is then checked for cross reactivity using an appropriate immunoassay. The serum is diluted to 2.5% (v/v) in buffer, with 0.08% sodium azide added as a preservative.

Frequently asked questions (FAQs)
What is the volume for one drop from the ReadyProbes dropper bottles?
The volume for one drop from the ReadyProbes dropper bottles is approximately 41 µL.
Can I use ReadyProbes 2.5% Normal Horse Serum (1X) as an antibody diluent for both primary and secondary antibodies?
Yes, ReadyProbes 2.5% Normal Horse Serum (1X) can be used as an antibody diluent for both primary and secondary antibodies, except for HRP-conjugated antibodies. The sodium azide added as a preservative is an inhibitor of horseradish peroxidase.
When would I use goat, horse, or chicken serum for blocking versus FBS or BSA?
For blocking, you should use a serum of the same or closely related species of the secondary antibody being used. If using a secondary antibody that was raised in goat or sheep, use goat serum (Cat. No. R37624 or 31873) for blocking; if using a secondary antibody raised in donkey or horse, use horse serum (Cat. No. R37625 or 31874) for blocking; if using a secondary antibody raised in chicken, use chicken serum (Cat. No. R37626) for blocking. FBS and BSA are alternatives when a specific species of serum is not available.
