ViewRNA Colorimetric Tissue assays are RNA in situ hybridization (ISH) assays for the reliable detection of mRNA targets within tissue sections at single-copy sensitivity. The ViewRNA assays use proprietary chemistry for the target-specific probe sets along with branched DNA (bDNA) technology to amplify target-specific signal and achieve superior signal-to-noise ratios compared to traditional colorimetric ISH signal amplification methods, which often amplify both the signal and background.

When combined with a ViewRNA Tissue probe set (purchased separately), the ViewRNA 1-Plex Kits enable the colorimetric detection of a single RNA target in tissue sections using the Fast Red, Fast Blue, or DAB substrate. ViewRNA 2-Plex Kits for the colorimetric detection of two RNA targets are also available in any combination of the three detection modes.

ViewRNA Tissue assays are RNA in situ hybridization (ISH) assays for the reliable detection of one or two mRNA targets within tissue sections. Visualize any gene in any tissue with single-copy sensitivity and no radioactivity. And in case your needs are not met by one of our 6500 catalog probe sets, we have decades of bioinformatics expertise and will design custom probe sets to any mRNA target of interest, free of charge.

With ViewRNA Tissue assays, you can achieve:
• Robust, single mRNA molecule detection in tissue sections
• Accuracy and specificity inherent to branched DNA amplification technologies
• Simultaneous detection of two RNA targets

The ViewRNA Tissue Assay is compatible with formalin-fixed, paraffin-embedded (FFPE) and cryopreserved tissue. Whereas traditional ISH technologies often amplify both signal and background, ViewRNA assays use branched DNA (bDNA) technology to amplify target-specific signal and achieve superior signal-to-noise ratios.

To meet various experimental needs, the ViewRNA Tissue assay is provided in a variety of kits and modules. The ViewRNA Tissue Core Kit and ViewRNA Modules can be purchased separately in different combinations to mix and match and create a colorimetric ViewRNA assay for detection of up to two RNA targets. For added convenience, ViewRNA 1-Plex and 2-Plex Kits containing the ViewRNA Tissue Core Kit and ViewRNA Modules can also be purchased, then supplemented with the ViewRNA Tissue Core Kit and ViewRNA Modules as needed for additional assays or substrates. The ViewRNA 1-Plex and 2-Plex Kits include sufficient reagents to stain 24 or 96 tissue sections. 

ViewRNA Fast Red 1-Plex Kit
The ViewRNA Fast Red 1-Plex Kit includes the ViewRNA Tissue Core Kit and ViewRNA Fast Red Module to detect a single mRNA target with red colored precipitate for ViewRNA colorimetric ISH tissue staining. The alkaline phosphatase (AP) substrate Fast Red is for use ViewRNA Type 1 or Type 6 Probe Sets (ViewRNA probe sets are sold separately).

ViewRNA Fast Blue 1-Plex Kit
The ViewRNA Fast Blue 1-Plex Kit includes the ViewRNA Tissue Core Kit and ViewRNA Fast Blue Module to detect a single mRNA target with blue colored precipitate for ViewRNA colorimetric ISH tissue staining. The alkaline phosphatase (AP) substrate Fast Blue is for use with ViewRNA Type 6 or Type 1 Probe Sets (ViewRNA probe sets are sold separately).

ViewRNA DAB 1-Plex Kit
The ViewRNA DAB 1-Plex Kit includes the ViewRNA Tissue Core Kit and ViewRNA DAB Module to detect a single mRNA target with brown colored staining for ViewRNA colorimetric ISH tissue staining. The horseradish peroxidase (HRP) substrate DAB is for use with ViewRNA Type 4 Probe Sets (ViewRNA probe sets are sold separately).

Frequently asked questions (FAQs)

How do ViewRNA assays compare to RNAScope assays?

ViewRNA and RNAScope technologies rely on the same signal amplification strategy - branched DNA amplification. Historically, both ViewRNA and RNAScope technologies originated from the same company, Panomics. The assays are expected to yield similar sensitivity and resolution, however each technology relies on its own set of proprietary reagents and probe set designs. Hence, the assays are not considered interchangeable or compatible. ViewRNA probe sets are not tested for RNAScope assaya and vice versa.

How is the signal amplified in ViewRNA assays?

The ViewRNA technology relies on branched DNA signal amplification strategy. Target probes complementary to the target transcript sequence are further hybridized with pre-amplifier, amplifier and label probes that consist of branched DNA, and form 'tree branches' that allow numerous label probes to attach. This approach allows higher signal amplification compared to traditional ISH techniques.

For ViewRNA assays, can I use the same set of wash solutions for all samples, including the positive and negative controls?

We do not recommend doing this. The negative control should be processed and washed separately from the rest of the samples. This is because the negative control does not contain any target probe sets and only the amplification reagents are added to it. If experimental samples are washed in the same beaker of wash solutions as the negative control, any unbound target probes that wash away can carry over to the negative control sample and cause unexpected positive signal (that will also appear to be very specific).
