Rapid Purification Protocol
The ChargeSwitch™ gDNA Normalized Buccal Cell Kit allows purification of genomic DNA from human buccal cell swabs or pelleted cells from a mouthwash in less than 15 minutes after lysates preparation. ChargeSwitch™ gDNA Normalized Buccal Cell Kit uses magnetic beads with a unique, ionizable functional group whose affinity for nucleic acids is pH-dependent to facilitate nucleic acid purification. High-quality genomic DNA is purified at a normalized concentration of 1-3 ng/μL in a total volume of 150 μL (Figure 1) by eliminating the need for lengthy quantitation and dilution steps. Samples can be stored for up to 2 weeks at 4°C before processing without a noticeable loss in DNA yield or quality.

Note: DNA yield varies and is dependent on several factors including the technique of the person taking the swab, whether the donor is a high or low shedder, and the type of swab used.

Validated for Single Tandem Repeat (STR) Profiling
After employing the ChargeSwitch™ gDNA Normalized Buccal Cell Kit, purified gDNA is suitable for use in STR analysis of poor or environmentally degraded samples, and for downstream applications such as PCR.

Compatible with Single Tube and High-Throughput Protocols
The ChargeSwitch™ gDNA Normalized Buccal Cell Kit is designed for automated processing of both small numbers of samples individually, as well as large numbers of samples using a liquid handling robot or a multi-channel pipette.

For processing up to 24 individual samples at a time the MagnaRack™ two-piece magnetic separation tube rack is required. This rack consists of a magnetic base station and a removable tube rack, which can hold up to 24 microcentrifuge tubes and can be used with most ChargeSwitch™ products.

For high-throughput applications utilizing 96 well blocks, the 96-Well Magnetic Separator is required. This rack can hold and separate up to 96 magnetic bead samples in a deep well plate when placed on top of the magnetic base station.

Frequently asked questions (FAQs)

I used water to elute my plasmid instead of the Elution Buffer with my ChargeSwitch kit, but saw very low plasmid yield. Why is this?

We strongly recommend using the Elution Buffer provided in the kit, and do not recommend elution with water. If you need to elute in any other buffer, be sure to use a buffer of pH 8.5-9.0 for efficient DNA elution.

What is the difference between the ChargeSwitch Pro kit and the Charge-Switch Pro filter kit?

The filter kit is based upon a dual nested column design, which facilitates the fastest and simplest protocol for obtaining high quality plasmid (less than 10 EU/µg DNA). Modified wash buffer 1 removes more endotoxin.

What is the typical yield when using the ChargeSwitch Pro Filter Plasmid Mini or Midi Kits?

The typical yield is 25 µg from a 5 mL culture for the ChargeSwitch Pro mini kit, 200 µg from a 25 mL culture for the midi kit, and 800 µg from a 100 mL culture from the maxi kit. The plasmid size range for the kits is typically 3-9 kb.

I want to isolate genomic DNA from buccal cells. How do I do this?

We would recommend using one of our MagMAX DNA Multi-Sample Ultra 2.0 Kit or ChargeSwitch gDNA Buccal Cell Kits, which were designed to isolate high-quality genomic DNA from buccal samples. We also offer normalized kits, which will normalize isolated DNA concentration from sample to sample.

How does ChargeSwitch technology work?

Based on a patented switchable chemistry, ChargeSwitch purification utilizes the charge on a solid surface for nucleic acid capture. At lower pH, the surface chemistry holds a positive charge and attracts nucleic acid. At higher pH, the surface chemistry is neutral and any bound nucleic acid is released.
