The Luminex FLEXMAP 3D Instrument System is the most advanced and versatile multiplexing platform. A fast read time, compatibility with both 96- and 384-well plates, and ease of integration with front-end automation systems make the Luminex FLEXMAP 3D system the platform of choice for high-throughput applications. Automated start up, shut down, and maintenance routines make the Luminex FLEXMAP 3D system easy to use and maintain. The system is compatible with ProcartaPlex multiplex immunoassays and QuantiGene Plex multiplex gene expression assays.

Features of the Luminex system include:

  • Multiplexing: up to 500 analytes in a single sample
  • Dynamic range (typical): ≥4.5 logs
  • Read time: 96-well plate in ∼20 min (up to 144,000 tests/hour); 384-well plate in ∼75 min (up to 153,600 tests/hour)
  • Daily start-up/shut-down: ≤30 min

Available options

  • Software: extra seat licenses, Basic Security Module, 21 CFR part 11 Module, Automation Module, LIS/LIMS Module
  • Service: service contracts, IQ/OQ

Luminex xMAP technology

The open architecture of Luminex xMAP technology uses imaging, microspheres, digital signal processing, and traditional chemistry—combining proven technologies in a unique way.

Advantages of Luminex xMAP technology include:

  • Helps reduce costs and labor through multiplexing
  • Smaller sample size requirements compared to single result assays
  • Enables fast, reproducible results from favorable kinetics of liquid bead array approach
  • Broad coverage of applications, including protein expression and gene expression profiling

How Luminex xMAP technology works

Sets of magnetic microspheres are color-coded with combinations of red and infrared fluorophores. Each bead set is coated with a reagent specific to a particular bioassay, allowing the capture and detection of several specific analytes from a single sample. Optics in the compact analyzer excite the internal dyes that identify each microsphere as well as any reporter dye captured, providing quantitation of individual analytes on each microsphere. The analyzer reads many beads from each set, enabling rapid and precise results for several targets within a single sample.

Frequently asked questions (FAQs)

How do the Luminex 200 Instrument System, Luminex FLEXMAP3D Instrument System and Luminex xMAP INTELLIFLEX System simultaneously monitor concentrations of several different analytes in a single sample?

Each protein detected requires a different bead region to differentiate between analytes. The Luminex 200 Instrument System, Luminex FLEXMAP3D Instrument System and Luminex xMAP INTELLIFLEX System are outfitted with two lasers. The first laser is a red diode laser that classifies beads (region) based on color and hence identifies which protein is being measured. The second laser is a green YAG laser that quantitates reporter fluorophore (R-Phycoerythin, RPE) associated with the bead surface and determines the amount of the protein that is bound to the bead. The beads pass through the system at a rate of several thousand per second, excited first by the red laser and then the green laser. The results obtained show protein identity and the intensity of the signal, respectively. The intensity of the signal is directly proportional to the concentration of the analyte captured in the sample. Therefore, by monitoring the spectral properties of the beads and the amount of associated RPE fluorescence, the concentration of one or more proteins can be determined.

What kits can ProcartaPlex Platinum Assay Buffer for Plasma Samples be used with?

ProcartaPlex Platinum Assay Buffer for Plasma Samples is designed to be used with the ProcartaPlex Platinum assay format. It is used for dilution of plasma samples and gives spike- and dilution-recovery results in the range of 70-130%. ProcartaPlex Platinum Assay Buffer for Plasma Samples is included in our Platinum ProcartaPlex Human Basic Kit for Plasma Samples (Cat.No. EPXP010-10421-901) and off-the-shelf Platinum panels. It is also offered as a stand-alone item (Cat. No. EPXP-11112-000).

What is the difference between xMAP Sheath Concentrate PLUS, RUO and xMAP Sheath Fluid Plus?

xMAP Sheath Concentrate PLUS, RUO (Cat. No. 4050023) is a 20X concentrated version of xMAP Sheath Fluid Plus (Cat. No. 4050021). It needs to be diluted before use.

xMAP Sheath Concentrate PLUS, RUO (Cat. No. 4050023) is a reformulation of xMAP Sheath Fluid Concentrate (Cat. No. 4050018). The formulation is updated from a single preservative to a mixture of three preservatives to allow for greater transit stability and to reduce the likelihood of future acquired microbial resistance.

Will there be any differences in performance of my instrument(s) when switching over to xMAP Sheath Fluid Plus from the original xMAP Sheath Fluid?

In-house testing has been done to confirm that assays deliver expected results when using xMAP Sheath Fluid Plus. White Paper is available upon request from Technical Support (techsupport@thermofisher.com).
