You can refer to page 9 of the AAV-MAX Helper-Free AAV Production System User Guide (https://assets.thermofisher.com/TFS-Assets/LSG/manuals/MAN0019619_AAV-MAX_Helper-Free_AAV_ProductionSystem_UG.pdf) for detailed Vi-CELL XR settings. We recommend diluting cell count samples with PBS to eliminate precipitate artifacts that may be created by interaction with the Vi-CELL reagents.

Does Viral Production Medium, AGT contain a surfactant? Do I need to add extra surfactant for culturing CTS VPC2.0 in bioreactors ?

While the full formulation for Viral Production Medium, AGT is proprietary information that cannot be disclosed, we can confirm that the medium contains a surfactant and that no additional surfactant would need to be added to the medium to support your suspension CTS VPC2.0 in 2 L bench-top bioreactor to 50 L bioreactor.

What is the concentration of glucose in Viral Production Medium, AGT?

Viral Production Medium, AGT is formulated to contain glucose at a concentration of 6 g/L.

What type of filter is recommended to prepare liquid aseptic media from Viral Production Medium, AGT powder?

Viral Production Medium, AGT powder needs to be reconstituted in deionized distilled water or Water For Injection (WFI) for Cell Culture (Cat. No. A12873) and then immediately sterilized by membrane filtration (positive pressure recommended). A low protein binding filter (such as PVDF) with a pore size of 0.2 µM is recommended.

How does Viral Production Medium, AGT powder differ from the research-grade Viral Production Medium (Cat. No. A48179)?

GlutaMAX Supplement (Cat. No. 35050061) is already present in the one-part Viral Production Medium, AGT powder.
