The MagMAX Viral/Pathogen Nucleic Acid Isolation Kit is designed to recover RNA and DNA from virus and gram-negative bacteria in samples such as blood, swabs, urine, and viral transport media (VTM). The kit utilizes MagMAX magnetic-bead technology, ensuring reproducible recovery of high-quality nucleic acid compatible with a broad range of applications, including real-time PCR, digital PCR, and next-generation sequencing.

Features of the MagMAX Viral/Pathogen Nucleic Acid Isolation Kit include:
• Automation-ready protocols designed for the KingFisher Flex and KingFisher Duo Prime systems
• Fast procedure allows for 96 samples to be processed in under 30 minutes
• Flexible protocol accommodates sample volume inputs from 200 μL to 2 mL of biofluids or transfer media
• No need for carrier RNA
• Elution volumes ranging from 50 to 100 μL

Maximum sample input in a 96-deep well or 24-deep well plate format
The optimized reagents included in the MagMAX Viral/Pathogen kit allow you to maximize the amount of sample input, thereby increasing the amount of RNA and/or DNA recovered. Up to 400 μL of sample can be processed per well in a KingFisher 96-deep well plate. For larger sample volumes, protocols are available to process up to 2 mL of sample per well in a KingFisher 24-deep well plate.

Benefits of MagMAX magnetic bead–based purification
Magnetic beads offer many benefits compared to other technologies for isolating nucleic acid. Beads bind the nucleic acid more efficiently than glass-fiber filters, resulting in higher and more consistent yields. Additionally, because filters and vacuum manifolds are not used, there is no risk of filter clogging due to cellular particulates in samples. This clogging issue is of particular concern with protein-rich, large-volume samples such as whole blood or plasma that are commonly used for viral testing.

The Dynabeads MyOne Silane contained in the MagMAX Viral/Pathogen Total Nucleic Acid Isolation Kit have an optimized silica-like surface chemistry with a highly specific surface area that allows efficient kinetics and high sensitivity in nucleic acid capture. These aspects make Dynabeads MyOne Silane ideal for the recovery viral RNA and DNA present at low concentrations in body fluids. Once captured, these nucleic acids can then be eluted in 50 μL of elution buffer for use in downstream applications.

Frequently asked questions (FAQs)

I accidentally stored my MagMAX Viral/Pathogen Nucleic Acid Isolation Kit at room temperature for several months. The beads are supposed to be stored at 4 degrees C. Are they still usable?

Yes, the kit is still usable.

I am getting MagMAX bead carryover in some wells of my plate. What might be causing this and how can I resolve it?

Some bead carryover is common and has not been documented to affect downstream assays. The most common cause of bead carryover is too much input sample. The best way to determine if the sample amount is the cause of the issue is to run PBS with Xeno (artificial DNA/RNA) in the lysis buffer and then check if there is still bead carryover. If that is not the cause, the initial sample may require special processing, such as bead beating or liquification. If there is still bead carryover without the original sample, and in particular, if the carryover is present in the same wells for multiple runs, then this may be an instrument alignment issue.

Do I need any additional consumables when using MagMAX kits on the MagMAX Express-96 instrument or a KingFisher instrument?

Plastic consumables need to be purchased separately in order to use the MagMAX reagents on the MagMAX Express-96 or a KingFisher instrument. The particular consumables and the amount required will vary by kit and protocol so please check the user guide for the particular MagMAX kit to be sure that all required plastic consumables are available prior to sample processing.

Where can I find Bindlt script protocol (.bdz) files for use of MagMAX kits with the KingFisher Flex Purification System and KingFisher Duo Prime Purification System?

The best place to download KingFisher Flex and KingFisher Duo Prime protocols for a particular MagMAX kit is the relevant MagMAX kit product page. The Bindlt script protocol (.bdz) files can be found under the Documents>Product literature section.

Can I scale up the MagMAX kit reaction volumes? If so, how should I do this?

Yes, reaction volumes for MagMAX kits can be scaled up. In general, the amount of MagMAX reagents should be scaled up proportionally for the increase in sample input amount. Please see the specific kit user guide for any recommendation. If none are included, then optimization may be necessary.
