Thermo Scientific Pierce DTT (Dithiothreitol), No-Weigh format, is offered as 7.7 mg x 48-tube microplates for convenient preparation of 100 μL of 0.5 M single-use stocks for addition to protein samples for effective reduction of disulfide bonds.

DTT is a popular reducing agent for many protein research applications, including preparing proteins for reducing polyacrylamide electrophoresis (SDS-PAGE) and reducing disulfide bonds for labeling sulfhydryl groups in conjugation reactions. At low concentrations, DTT is used to stabilize proteins and enzymes with free sulfhydryl groups to keep monothiols in a reduced state. DTT reduction is also used to prepare proteins for mass spectrometry.

Properties of DTT:

Synonyms—1,4-dithiothreitol; DTT; Cleland’s Reagent; 1,4-dimercapto-2,3-butanediol
Molecular weight (MW)—154.24
Molecular formula—C4H10O2S2
CAS number—3483-12-3

DTT is not stable in solution and should be prepared fresh for each use. The No-Weigh format consists of a 48-microtube tray containing dry, stable, single-use aliquots of the DTT chemical to eliminate the need to weigh small amounts of reagent and prevent waste of unused DTT. Just puncture the seal of one microtube, add 100 μL of water or buffer, and mix to dissolve. In just a few seconds you will have a solution of 500 mM DTT, ready for a variety of laboratory methods. Dilute as necessary or add the appropriate volume of the 500 mM stock solution to achieve the amount of DTT reducing agent required for your specific application.

Features of the No-Weigh format:

Saves time—just add 100 μL water or buffer to make 500 mM DTT solution in seconds
Eliminates waste—makes 100 μL at a time, just the right amount for most applications
Always fresh—ensures full reducing strength for every use; no need to store stock solutions

With the convenient No-Weigh format, a 500 mM DTT solution can be made in less than 30 seconds. The unique packaging ensures that the DTT reducing agent is at full strength and able to protect proteins from oxidative damage or reduce any disulfides prior to electrophoresis.

For Research Use Only. Not for use in diagnostic procedures.

Frequently asked questions (FAQs)

What is the shelf life of Pierce DTT (Dithiothreitol), No-Weigh Format (Cat. No. A39255)?

Pierce DTT (Dithiothreitol), No-Weigh Format is covered under our general 1-year warranty and is guaranteed to be fully functional for 12 months from the date of shipment, if stored as recommended. Please see section 8.1 of our Terms & Conditions of Sale for more details.

Can you please recommend a method for reduction, alkylation, and digestion of an antibody?

Prepare 1 mg/mL antibody in 7.0 M Guanidine HCl, 100 mM Tris, pH 8.3. For reduction, add DTT (Cat. Nos. A39255, 20290) solution at 10 mM final concentration and incubate for 30 mins at room temperature. For alkylation, add sodium iodoacetate at 20 mM final concentration and incubate in the dark for 20 mins at room temperature. Perform buffer exchange followed by digestion using Pierce MS grade Trypsin, Cat. Nos. 90058, 90057 (1:10 enzyme to substrate ratio).

How does a product with "No-Weigh" format reduce my concern over reagent stability?

The No-Weigh product format, where small amounts of reagent are pre-weighed and packaged separately, prevents the contamination and loss of reagent reactivity over time associated with repetitive opening and closing of a single vial. This format enables use of a fresh vial of reagent each time and eliminates the hassle of weighing out small amounts of reagent.
