Media Preparation

Powdered media is dissolved in Type I water, pH is adjusted, followed by filter sterilization. Stirring and filtration can be performed in biological safety cabinets. Alternatively, ready-made liquid media can be used.

Cell Revival

Cells are removed from long-term storage and quickly brought to 37°C. They are washed to remove the cryopreservation medium, plated in fresh medium.

Adherent Cells Growth

This is anchorage-dependent cell culture, where cells are grown on the bottom of a tissue culture flask in a medium.

Scale Up

Involves the transfer of cell culture system from laboratory scale to industrial scale which requires proper media supplement, culture ware and antibiotics.

Passage

Spent media is removed from the cells and washed with buffers. If needed, they are treated with dissociation reagents and washed again. A portion of cells is reseeded onto new culture ware.

Transfection

DNA and RNA are introduced in cells using transfection reagents to produce recombinant proteins, or to enhance or inhibit gene expression

Characterization & Analysis

Cells may be fixed and/or stained for visualization, can be lysed for protein purification, genetic analysis or other molecular biology studies. They may also be used for flow cytometry

Harvest

Cells are washed and centrifuged, and their population is checked by a cell counter and EVOS microscope.

Cryopreservation & Storage

After washing and centrifuging, they are resuspended in a storage medium in a cryovial. After being cooled to -80°C, they are stored in liquid N2 and can be preserved for years.
