PA3-014 detects human, canine and porcine, rat and mouse sec61 alpha from microsomes and shows no cross-reactivity with other sec61 isoforms.
PA3-014 has been successfully used in Western blot procedures. By Western blot, this antibody detects an ~38 kDa protein representing sec61 alpha from pig microsomes.
The PA3-014 immunogen is a synthetic peptide corresponding to residues C(13) V I L P E I Q K P E R K I Q F K E K(31) of canine sec61 alpha. This sequence is completely conserved between human, mouse, rat, and canine.
The Sec61 complex is responsible for translocation of proteins across the rough endoplasmic reticulum membrane. Sec61 is composed of three subunits: alpha, beta, and gamma; that associate to form a heterotrimer. Multiple heterotrimers associate forming the ER translocon. Sec61 alpha is a polytopic protein that has ten membrane-spanning loops. Sec61 has been found to be tightly associated with membrane bound ribosomes, either directly or through adaptor proteins, and is required for assembly of membrane and secretory proteins.
