Reconstitute the lyophilized antiserum by adding 1 mL sterile distilled water.
Inter-species cross reactivity is a normal feature of antibodies to Serum proteins, since homologous proteins of different species frequently share antigenic determinants. Crossreactivity of this antiSerum has not been tested in detail, however a reaction has been observed with Human albumin, Chicken ovalbuminDNP, but not with native Chicken ovalbumin.
In immunoelectrophoresis use 2 µL serum or equivalent or DNP-conjugates against 120 µL antiserum. In double radial immunodiffusion use a rosette arrangement with 10 µL antiserum in 3 mm diameter centre well and 2 µL serum samples (neat and serially diluted) in 2 mm diameter peripheral wells. In single radial immunodiffusion (Mancini) use 0.5 to 1% antiserum in the agar gel.
Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at 4°C, not refrozen, and preferably used the same day.
Dinitrophenyl is any chemical compound containing two nitro functional groups attached to a phenyl ring. It is a hapten used in vaccine preparation. Dinitrophenyl does not elicit any immune response on its own and it does not bind to any antigen. Albumin is a soluble protein which constitutes approximately 50% of blood serum protein, and acts as a regulator of blood colloidal osmotic pressure and blood volume. Albumin is also an important transporter of substances within the bloodstream, such as fatty acids, thyroid hormones, metal ions and steroids.
