Immunohistochemistry (PFA fixed): incubate antibody for 30 min at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris Buffer with 1mM EDTA, pH 9.0, for 45 min at 95 degrees C followed by cooling at RT for 20 minutes.
Mucins are high molecular eight glycoproteins with 80% carbohydrate contents and the remaining 20% is constituted by protein core. Gastric mucin is found in normal human gastrointestinal tract. Resurgence of gastric mucin reactivity during colonic carcinogenesis is due to re-expression of the peptide core of gastric (or fetal colonic) mucins.
